How a flow cytometer works?

In this second article, we will delve into the operational details of this technology and explain why it stands as an all-encompassing solution for your laboratory needs. 


How does it work?

Now that we understand the essential components of a flow cytometer, let's explore how it operates to analyze spermatozoa efficiently. Here are the main steps in the process:


 

1. Fluorescence Incubation: Before analysis, the sample is often treated with fluorescent dyes that bind to specific cellular components. These dyes emit fluorescence when exposed to laser light.

 

2. Laser Excitation: The sample is injected into the flow cytometer's capillary system. The laser, usually emitting light at 488 nm, is directed at the flowing particles. When the laser beam hits a fluorescently labeled particle, it excites the dye molecules, causing them to emit fluorescent light.

 

3. Detection Channels: Inside the flow cytometer, different receptors are used to capture the emitted light. These receptors are divided into channels, including the Forward Scatter Channel (FSC) for analyzing cell size and the Side Scatter Channel (SSC) for evaluating cell granularity. Additionally, various receptors are dedicated to analyzing the fluorescence emitted by the dyes, allowing for a detailed characterization of the semen sample.

 

By collecting data from these detectors, the flow cytometer generates a multi-dimensional profile of the spermatozoa in the sample, offering valuable information.
